1412 “Telescope model” for coordinated organ morphogenesis and stem cell formation

Although hair follicle stem cells are central to the homeostasis and regeneration of adult follicles, their developmental process had not been fully elucidated. Here, by combining marker-independent long-term three-dimensional (3D) live imaging single-cell transcriptomics, we provided dynamic four-dimensional (4D) atlas with cellular dynamics, cell lineages, transcriptome changes in entire epithelium developing mouse follicles. Our 4D demonstrated that follicles divided into cylindrical compartments, these compartments exist as concentric ring-shaped pre-patterns placode, future derived from peripheral ring compartment but previously suggested suprabasal cells. Given two-dimensional rings placode transforming a series resembles extension telescope, call this “telescope model”. The telescope model may explain spatiotemporal coordination tissue morphogenesis fate specification during formation 3D consisting various epithelial one which is lineage.
